What standard certificates omit

Most standard certificates stop at basic identity and a nominal purity figure. Rigorous incoming quality control demands tests frequently omitted due to cost:

  • Endotoxin (LAL): bacterial endotoxin load.
  • Heavy metals: lead, arsenic, cadmium, mercury.
  • Residual solvents: process solvents remaining from manufacture.
  • Impurity profiling: identification of secondary compounds.
  • Content assay: exact quantification of the labeled peptide’s mass.
  • Water content: residual moisture impacting mass formulation.
  • Counterion / salt form: specific salt state (acetate, TFA, HCl).
  • Bioburden / sterility: assessment of microbial load.

Why paperwork alone does not hold

FDA and cGMP (21 CFR 211.84)

Quarantine is the default state for received material. The FDA mandates at least one specific confirmatory identity test on every incoming lot. A supplier’s COA cannot be substituted for independent receiving analysis without established validation. Read 21 CFR 211.84 at eCFR.

Empirical evidence (JAMA, 2017)

Independent analysis of 44 unapproved drug products sold online showed the paperwork failed to reflect physical reality 48% of the time, highlighting the statistical risk of bypassing independent testing. JAMA. 2017;318(20):2004–2010.

Document fraud & forgery

Independent testing facilities document widespread forgery of COAs by raw material suppliers. A static PDF holds no analytical weight unless verifiable via an accession number on the third-party laboratory’s secure server.

Pharmacopeial standard (USP <1083>)

USP standards forbid relying on a supplier’s COA for critical quality attributes unless the buyer has performed concurrent, independent laboratory testing on multiple lots to prove consistent accuracy.
